the application prospects of real-time fluorescence quantitative PCR in medical detection and various other fields are very broad.


Technical Introduction



 RT-QPCR refers to the addition of fluorescent dye SYBR or specific probes labeled with fluorescence in the PCR reaction system, and the real-time online monitoring of the PCR reaction process using the accumulation of fluorescence signals. 

Due to the linear relationship between the Ct value of the PCR amplified gene and the initial copy number of the template, it becomes a quantitative basis and can achieve relative or absolute quantification of the target gene in the sample.

Fluorescent quantitative PCR not only achieves absolute quantification, but also has characteristics such as sensitivity and specificity, high energy for multiple reactions, high automation, pollution-free, real-time, and accuracy.




Method introduction


According to Real time qPCR, there are two types of fluorescent chemicals used: fluorescent dyes and fluorescent probes. 

Depending on the different fluorescent substances used,

 the commonly used methods for this technology include SYBR Green dye method and TaqMan probe method



Operation method


· Design specific primers or fluorescent probes based on the target gene sequence;

· Extract DNA/RNA, RNA reverse transcription, electrophoresis, and concentration measurement from each sample;

· Degenomic DNA, RNA reverse transcription;

· Configure a PCR reaction system and add the necessary solutions for templates, primers, dyes, and dNTP systems;

· RealTime PCR amplification;

· Obtain offline data and analyze amplification curves, dissolution curves, and expression differences.
